Scientists reverse age-related vision loss, eye damage from glaucoma in mice — ScienceDaily

Harvard Medical School scientists have successfully restored vision in mice by turning back the clock on aged eye cells in the retina to recapture youthful gene function.

The team’s work, described Dec. 2 in Nature, represents the first demonstration that it may be possible to safely reprogram complex tissues, such as the nerve cells of the eye, to an earlier age.

In addition to resetting the cells’ aging clock, the researchers successfully reversed vision loss in animals with a condition mimicking human glaucoma, a leading cause of blindness around the world.

The achievement represents the first successful attempt to reverse glaucoma-induced vision loss, rather than merely stem its progression, the team said. If replicated through further studies, the approach could pave the way for therapies to promote tissue repair across various organs and reverse aging and age-related diseases in humans.

Just a few doses of an experimental drug can reverse age-related declines in memory and mental flexibility in mice, according to a new study by UC San Francisco scientists. The drug, called ISRIB, has already been shown in laboratory studies to restore memory function months after traumatic brain injury (TBI), reverse cognitive impairments in Down Syndrome, prevent noise-related hearing loss, fight certain types of prostate cancer, and even enhance cognition in healthy animals.

In the new study, published December 1, 2020 in the open-access journal eLife, researchers showed rapid restoration of youthful cognitive abilities in aged mice, accompanied by a rejuvenation of brain and immune cells that could help explain improvements in brain function.

Engineered immune cells elicit broad response to HIV in mice, offering hope for vaccine — ScienceDaily

Unlike so many other deadly viruses, HIV still lacks a vaccine. The virus — which continues to infect millions around the world — has proven especially tricky to prevent with conventional antibodies, in part because it evolves so rapidly in the body. Any solution would require coaxing the body into producing a special type of antibody that can act broadly to defeat multiple strains of the virus at once.

This week, scientists at Scripps Research moved closer to attaining that holy grail of HIV research with a new vaccine approach that would rely on genetically engineered immune cells from the patient’s body.

In experiments involving mice, the approach successfully induced broadly neutralizing antibodies — also called bnabs — that can prevent HIV infection, says principal investigator James Voss, PhD, of Scripps Research. The study appears in Nature Communications.

A combination of innate differences and diet-induced changes to the reward system may predispose some mice to overeat, according to research recently published in JNeurosci.

Food is fuel, but the rising levels of sugar and fat in modern diets make the brain treat it as a reward. One brain region called the ventral pallidum (VP) serves as a hub between reward areas and the hypothalamus, a region involved in feeding behavior. Intertwining food and reward can lead to overeating and may be a contributing factor to diet-induced obesity.

Gendelis, Inbar, et al. measured electrical activity in the VP of mice with unlimited access to high fat, high sugar food for several months. Eating the unhealthy diet changed the electrical properties of VP neurons: the membrane voltage and firing rate decreased, making it harder for neurons to send messages to each other.

In mice, cadmium exposure during pregnancy linked to obesity in female offspring — ScienceDaily

In a mouse study aimed at modeling human exposure to the toxic metal cadmium, researchers from North Carolina State University found that female offspring of mice exposed to cadmium during pregnancy became obese in adulthood, developed fatty livers and could not process glucose normally. Male offspring were not affected in the same way. The study also sheds light on how cadmium exposure could affect mitochondrial function and developmental signaling pathways in the liver.

Cadmium is a toxic heavy metal. Environmental exposure commonly occurs from fossil fuel emissions, smoking, and some food and water sources. It is an accumulative toxin, meaning that at normal exposure levels, exposure must occur over years to produce toxic effects. However, there is compelling evidence in humans that relatively short-term developmental exposure to cadmium may increase risk for obesity and obesity-related conditions such as cardiovascular disease and type 2 diabetes.
